Combinatorial therapy allows viruses to destroy tumors

For several years, researchers have been developing a new approach to treating cancer that uses viruses to infect and kill cancer cells while leaving normal cells unharmed. Recent data have indicated that this approach, which is known as oncolytic virotherapy, has potential. Now, Richard Vile and colleagues, at the Mayo Clinic, Rochester, have found that this approach can be combined with a standard clinical therapy to provide substantial regression and cure of tumors in mice, leading them to suggest that this combinatorial approach could be of tremendous benefit in the clinic.

Scientists funded by the Biotechnology and Biological Sciences Research Council (BBSRC) at the University of Birmingham have discovered that a gene called DAF-16 is strongly involved in determining the rate of ageing and average lifespan of the laboratory worm Caenorhabditis elegans (C. elegans) and its close evolutionary cousins. DAF-16 is found in many other animals, including humans. It is possible that this knowledge could open up new avenues for altering ageing, immunity and resistance to stresses in humans. The research is published today (01 April) in PLoS ONE.

Results reported in the April 2nd issue of the journal Cell, a Cell Press publication, reveal a new source for the drug resistance that crops up all too often and quickly in the tumors of cancer patients undergoing therapy. First the bad news: all cancer cells might have the capacity to enter a drug-tolerant state. But there’s some potentially very good news too: in some cases there may be a way to reverse or block cancer’s drug resistance.
